Finding a New Prognostic Biomarker for Metastatic Colorectal Cancer

چکیده
cer progression. However, for these results to be adopted in the clinical filed, it is very important that a large-sample-size study be brought conducted in a validation setting. The findings of this research finding suggest that PlGF may constitute a novel approach to colorectal cancer treatment. Several agents against PlGF have already developed or are currently under development. Among these agents, two novel agents against PlGF will be briefly discussed. Firstly, aflibercept (ZALTRAP) is a recombinant fusion protein that acts as a soluble receptor that binds to VEGF-A, VEGF-B and PIGF. In April 2011, Van Cutsem et al. [3] reported that aflibercept improved the primary endpoint of overall survival in the Velour phase III clinical trial for second-line treatment for metastatic colorectal cancer. Secondly, the humanized anti-PlGF mAb (TB-403) is directed against PlGF. A trial with TB-403 has only completed its first phase. Antitumor activity has been demonstrated with TB-403 in human tumor xenograft models of renal cell carcinomas and hepatocellular carcinomas. The mechanism of action for TB-403 is currently under investigation; preclinical studies suggest its effects may include blocking tumor angiogenesis and primary tumor growth, as well as inhibition of metastasis. In addition, PIGF inhibition may complement and potentiate the antitumor effects of VEGF inhibition, in part by inhibiting macrophage recruitment [4].

Background: Kirsten rat sarcoma (KRAS) gene is a target of genetic alterations which are diagnostic and prognostic biomarkers in patients with metastatic colorectal cancer who are treated with monoclonal anti-EGFR antibodies such as cetuximab and panitumumab. KRAS mutations are seen in 35-42% of patients with colorectal cancer. The high frequency of these mutations in colorectal cancer represen...
متن کاملSNHG6 203 and SNHG6 201 Transcripts Can be Used as Contributory Factors for a Well-Timed Prognosis and Diagnosis of Colorectal Cancer
Background:Long non-coding RNAs, as a big part of non-coding RNAs, are considered functionally more than past. These transcripts could be involved in carcinogenesis. SNHG6, as a long non-coding RNA, has been reported to be expressed more in colorectal cancer tissues than non-cancerous ones. Colorectal cancer as a malignancy needs fast prognostic and diagnostic methods for well...
متن کاملبررسی بیان miRNA-21 در سرطان کولورکتال
Background: Colorectal cancer is the third most common cancer in the world. Non-coding RNA especially miRNAs have important regulatory roles in cancer. miRNAs are small non coding RNA 21-23 nucleotides long which have different levels of expression between tumors and normal tissues. This study was designed to compare expression level of miRNA-21 between Iranian population colorectal cancer tiss...
متن کاملA Pilot Study of CK19, CK20 and GCC mRNA in the Peripheral Blood as a Colorectal Cancer Biomarker Panel
Colorectal cancer remains one of the major cancer- related deaths despite progress in the treatment during past decades. Detection of disease at earlier stages reduces its mortality. The aim of current study was to investigate expression of Cytokeratin 19 (CK19), Cytokeratin 20 (CK20) and Guanylyl Cyclase C (GCC) mRNA in peripheral blood of non- metastatic colorectal cancer patients which may r...

Colorectal cancer (CRC) is one of the most common types of cancers worldwide. Despite the improved therapeutic and diagnostic strategies and screening programs, morbidity and mortality of CRC is still considerable. Currently, most common approaches for diagnosis of CRC are colonoscopy and fecal occult blood test (FOBT). Because of the invasive nature of colonoscopy and low sensitivity of FOBT, ...
متن کاملLong Non-coding RNA ZEB1-AS1 Promotes Tumorigenesis and Metastasis in Colorectal Cancer
Emerging evidence implicates that a large fraction of human genome was transcribed but the transcripts known as long non coding RNA are not translated into proteins. They are contributing in different cellular processes, including cellular proliferation and apoptosis. LncRNAs were found to play critical roles in many diseases and act as key regulators in malignancies. In this study, we investig...
